Data evaluation Template quality is a key contributor to the quality of the resultant sequence data. It is preferred that all DNA templates are post-extraction purified using a column-based, DNA purification kit. This ensures that residual RNA, salt, protein and other contaminating chemicals do not affect the quality of resolution (quality of base calls) or signal intensity of the sequence data. Alternatively, the modified alkaline lysis PEG precipitation method produces high quality template.
Up to 1000 bases can be sequenced per run, however, the accurate read length is largely dependent upon a variety of factors: Quality of the template >> read more Quantity of the template >> read more Primer design >> read more Removal of residual dyes post cycle sequencing >> read more Trouble-shooting resources Sequence data evaluation and trouble-shooting guide (pdf) Automated DNA sequencing chemistry guide (pdf) BigDye terminator v3.1 cycle sequencing kit protocol (pdf) |
